NG+ bosses kick your ass hard, and when I started an ascended playthrough, with all the beads and prayers already collected and knowing all the enemy patterns, I still struggled with them - and that's with using the flasks. Can't imagine the effort it'd take without the flasks.
Are you sure about this? I was under the impression that you have to beat the bosses in a single playthrough.
I am not sure about this, cause I saw the same, but on the other hand saw someone on a forum who mentioned that missed the procedure on the third boss and after going to NG+ and killing the third boss got the achiemevent (bottom comment):

I started using the noxious gas one when I got to Crisanta since she recovers too fast to hit her with most prayers and that thing will melt anyone you stand near. I first tried every Amanecida using it and even killed the last one so fast I skipped the final phase
