In the Devil May Cry novel Gilver said that if he cuts out Dante's heart, he wouldn't be able to recover, no matter how much demon blood runs through his veins.
I have to point out that the novel is semi-canon, but I guess this statement has a hint of truth. Hideki Kamiya also said that the heart is the most important, so this might be the case, even more that Vergil was "dead" when Mundus removed his human heart.
I also think that their regenerative abilities have their limits when they are exhausted.
About Sparda; it seems that he didn't die completely, honestly it's hard to tell, because in Nico's reports it is written that Sparda sealed himself~ this isn't quite equal to death, although I can't really tell, because every source says differently.
Moreover, we've seen many times that Vergil and Dante were stabbed in the chest with a sword and nothing serious happened to them, but "cutting out" and "stabbing" can have a completely different effect on them and maybe they can't regenerate after that, but who really knows, with them everything is possible, we can only speculate about how and who can kill them.
Apart from that, unfortunately I don't remember Dante's statement from DMC4 that you are referring to, but I assume he meant about human strong fighting spirit and strong will, or rather all the traits that distinguish humans from demons and that give humans an advantage over them.
Humans aren't weak at all in this universe, because if you look at Lady, how the hell she can move and kill demons that could certainly be faster, stronger and way more resistant than any normal human being? Sure, she can't beat a Dante or Vergil, but that doesn't mean she's weak compared to many of the demons we face in DMC 3. You could say there's little demonic power being used when Dante didn't awake Rebellion yet, or when Nero lost his arm.
Both humans and demons in DMC were created at the same time, one can get stronger through the other one, that's why Dante and Vergil are insanely powerful, besides being the Sons of Sparda. Think about it, they are already getting pretty close to their father's full strength, and i doubt Sparda lived less than 3 digits when living in hell. His sons gonna keep growing way past his prime.
Vergil did die back in DMC 1, his soul was probably sent to hell and his body regenerated, but not enough to survive out of it (before the Urizen/V stuff.) That's probably what could have happened to Sparda, not having enough power to maintain his body and live longer, especially out of his world.
Humans can't withstand hell, they are corrupted by it and are driven mad, DMC novel stuff. Demons could get negative effects If they stay too long in the human world too, who knows.
